Harvard University Study Finds Benzyl Butyl Phthalate (BBP) Damages DNA and Causes Germline Dysfunction
Researchers from Harvard Medical School have published a study in PLOS Genetics revealing that the commonly used plasticizer, benzyl butyl phthalate (BBP), can damage DNA and lead to dysfunction in germ cells.
On October 24, 2024, researchers from Harvard Medical School published a paper titled Exposure to Benzyl Butyl Phthalate (BBP) Leads to Increased Double-Strand Break Formation and Germline Dysfunction in Caenorhabditis elegans* in PLOS Genetics. This new study highlights the potential harm BBP can cause to the reproductive system.
The research team tested various doses of BBP on Caenorhabditis elegans (C. elegans), a model organism, and found that a 10μM dose of BBP had the most significant impact on germ cells. This exposure resulted in increased apoptosis of germ cells and defects in chromosome organization. Specifically, BBP caused oxidative stress and DNA double-strand breaks, leading to cell death and oocytes with chromosomal abnormalities.
Mass spectrometry analysis showed that C. elegans could metabolize BBP into mono-butyl phthalate (MBP) and mono-benzyl phthalate (MBzP). The levels of BBP, MBP, and MBzP detected in C. elegans were similar to the ranges found in human biological samples, such as serum and urine. This indicates that C. elegans is a valuable model for studying the effects of BBP on human health.
This study emphasizes the toxic effects of BBP, a common plasticizer, and its potential harm to the reproductive system.
Other studies have also highlighted the negative impacts of plasticizers on reproductive health. For instance, di(2-ethylhexyl) phthalate (DEHP) has been shown to cause excessive DNA damage during the reproductive process in C. elegans, interfering with the repair system and leading to defects in oocyte formation and early embryonic development. DEHP is one of the most widely used plasticizers, with a global annual production of up to 4 million tons. People are exposed to DEHP through various products, including toys, clothing, food packaging, medical devices, and personal care items.
Hazards of Phthalates:
Toxicity to the Male Reproductive System: Phthalates are known to cause testicular toxicity, including severe testicular atrophy, significant changes in testosterone levels, a reduction in spermatogenic cells, a rapid decrease in testicular enzyme activity, and a reduction in sperm count.
Toxicity to the Female Reproductive System: Phthalate esters have toxic effects on the uterus and ovaries, potentially leading to extended estrous cycles, reduced progesterone levels, uterine weight gain, and altered ovulation cycles, including delayed or anovulatory cycles.
Effects on the Blood System: Exposure to phthalates can result in a significant reduction in hematocrit, decreased hemoglobin levels, and increased platelet adhesion.
Teratogenicity: Research suggests that exposure to low doses of phthalates can significantly increase the probability of fetal death. Acute exposure may also lead to cardiac, renal, and pulmonary toxicity, potentially resulting in death.
Hepatic Toxicity: Phthalates can cause oxidative damage to proteins, liver cell membranes, and DNA within cells, potentially even damaging stem cells.
Carcinogenicity: Long-term exposure to phthalates greatly increases the likelihood of developing malignant stem cell tumors.
